          Abstract :
          The present work comprises three major chapters, which begin with an introductory part
          and a literature survey.
          In the present invention, mucoadhesive in situgel-based systems has emerged asa new
          approach  to  drug  delivery  system.In  situ,gel-forming  systems  are  viscous  liquids  that
          undergo to a gel phase upon exposure to physiological conditions. The formation of the
          gel  depends  on  the  factors  such  as  temperature  fluctuation,  pH  changes,  and  ion
          exposure  from  which  the  drug  is  released  continuously  and  in  a  controlled
          manner.Controlled  release  of  drugs  offers  the  benefit  of  reduced  administration
          frequency and improved patient compliance. The dose of the drug is reduced which is
          why the toxicity, is compared to conventional treatment.

          In the present research work, an attempt has been made to formulate metronidazole and
          clotrimazole  in  situ  gel  for  the  effective  management  of  periodontitis,  and  vaginal
          candidiasis,  &  otomycosis  with  local  delivery  for  a  controlled  release.  For  this  study,
          various  concentrations  of  Poloxamer  407,  Poloxamer  188,  carbopol  974,  gellan  gum,
          HPMC E50, Natrosol 250, and Klucel HF polymers were used. The formulated in situ gel
          was  evaluated  for  clarity,  in  vitro  gelling  capacity,  gelation  time,  pH,  viscosity,  gelation
          temperature, % drug content, spreadability, in vitro drug release, and in vitro antimicrobial
          study. 32 full factorial designs and desirability functions were applied. All the formulations
          were  developed  using  different  concentrations  of  the  above  polymers.  The  developed
          formulations  showed  satisfactory  results  for  different  evaluation  parameters.  We  had
          considered the best formulation based on desirability and overlay plot.
